Transgenic luminescent PhotoTopo® mice

Axxam scientists have a developed a novel transgenic mouse model animal for applications in discovery research and cellular screening. We have stably expressed our proprietary photoprotein Photina®, in a mouse embryonic stem cell (mES) line as a reporter gene, and this mES-reporter cell line was used to generate a transgenic mouse expressing the photoprotein reporter gene driven by an ubiquitous promoter, with most tissues shown to express the calcium sensitive reporter protein.

This unique whole animal model can be used to prepare tools for screening in HTS applications such as:

  • primary cells from numerous tissues sources
  • mouse Embryonic Stem cells for differentiation into a variety of tissue types
  • adult stem cells

Primary cells derived from the transgenic animal tissues expressing Photina® can be isolated, cultivated and used directly for functional experiments of target-induced intracellular calcium movements. In addition these cells can be used as a true control of the stem cell derived primary cells of the same specific lineage.

The transgenic Photina® mice can also be interesting for crossing with some pharmacologically relevant mutant mice and for photo-imaging activities.
