The chAMPion cellular reporter system is a novel combination of Axxam’s proprietary photoprotein technology coupled with the sensitivity of using cyclic nucleotide gated channels for measurement of cAMP levels.

The detection chemistry is luminescent in nature, and therefore affords all the advantages of this detection modality, such as extremely low background, exquisite sensitivity and a greatly reduced incidence of compound interference. chAMPion represents a platform that permits interrogation of all the major classes of coupled G-protein coupled receptors, Gq, Gi and Gs, as the adenylate cylcase/cAMP modulated pathway is also linked to calcium flux via the CNG sensor, and the Gq/PLC pathway naturally induces changes in calcium levels, both of which can be monitored by activation of the calcium sensitive photoprotein light production.

The chAMPion cell line display many advantages: they represent a real-time assay for sensitive detection of cGMP and cAMP, are suitable for HTS and uHTS, require short incubation times decreasing the false positives rate, avoid any fluorescence interference from test compounds, enable the characterization of GPCRs coupled to all the different Gα proteins, and represent an assay platform for different multiple target classes, such as: cGMP: Guanylate Cyclases, cGMP-PDEs, NO Synthases, Adenylate Cyclases and cAMP-PDEs
