Choosing the Axxam Screening Technology group for your high-throughput screening needs is about more than just ‘hits’. It’s about variable library capacity, tight coupling with our assay development scientists, both cell-based and biochemical assays, using luminescent and fluorescent detection - and all using the latest instrumentation and generating high-quality analytical reports.
We provide high-throughput screening of the Axxam
compound libraries or
chemical libraries that you supply to us - from small and narrowly-focused libraries
to large and broadly-focused. And you can benefit from the tight coupling between
assay development and screening at Axxam. First, your assays can move seamlessly
from assay development into HTS with minimal delay if you also partner with Axxam
for assay development. Second, optimal performance of the screening campaign
is ensured with scientists from both groups committed to the success of the complete
project.
While our focus is on cell-based functional assays, using luminescent and fluorescent readouts, we also can process a variety of biochemical assays, depending on the specific requirements. This means we can screen most targets relevant to the pharmaceutical industry, including GPCRs, ion channels, and enzymes.
The flexible screening facility we have created can adapt to the demands of multiple assay formats. Our main screening platform is built around a CyBio® screening system. For luminescent assays we employ a CyBio® Lumax
HTS - a highly-sensitive luminescent plate reader. For fluorescent assays, we
have integrated the Molecular Devices FLIPRTETRA, permitting a wide range of excitation/emission
options. We also utilize more sophisticated fluorescence-based plate readers
for particular assays if advantageous. And for cherry-picking, we have a completely
automated system that allows easy retesting of putative hit compounds. We typically
employ 384-MTPs for all steps, although we can utilize 96- and 1536-MTPs when
necessary.
At the end of the screening campaign, we’ll be providing more than ‘hits’. Our emphasis is on high-quality
data. That means attention to every detail during the complete project - careful choices of readout technologies, proper concentrations for reference ligands, effective counter-screens and more. Even if you provide a fully-configured assay, our experienced assay development scientists can offer fine adjustments to increase the quality of your hits.
All screening data are automatically deposited into our custom relational database. Automated real-time data processing allows us to retest within the same day those plates that do not meet our strict quality-control parameters for Z’, hit distribution and failed wells. Sophisticated data processing methodologies monitor for systematic errors. We want you to be confident of the quality of your hits, so we retest all putative hits by multi-well hit confirmation experiments. And when it comes to reporting data back to you, you can choose from a variety of options including simple text or Excel tables, database exports, and graphical views using Spotfire® visualizations.
Compounds collection (Libraries)
Axxam high-throughput screening services offers three different options for library screening:
- Your Library – we screen compound collections that you supply.
- Axxam Synthetic Compound Collection – we screen our
library composed of 100,000 different synthetic compounds.
- Natural Compound Collection – we screen a collection of 100.000 different natural compounds available to Axxam from one of our partners.
